  1. The performance of different propensity score methods for estimating marginal hazard ratios

    Propensity score methods are increasingly being used to reduce or minimize the effects of confounding when estimating the effects of treatments, exposures, or interventions when using observational or non-randomized data. Under the assumption of no unmeasured confounders, previous research has shown that propensity score methods allow for unbiased estimation of linear treatment effects (e.g., differences in means or proportions). However, in biomedical research, time-to-event outcomes occur frequently. There is a paucity of research into the performance of different propensity score methods for estimating the effect of treatment on time-to-event outcomes. Furthermore, propensity score methods allow for the estimation of marginal or population-average treatment effects. We conducted an extensive series of Monte Carlo simulations to examine the performance of propensity score matching (1:1 greedy nearest-neighbor matching within propensity score calipers), stratification on the propensity score, inverse probability of treatment weighting (IPTW) using the propensity score, and covariate adjustment using the propensity score to estimate marginal hazard ratios. We found that both propensity score matching and IPTW using the propensity score allow for the estimation of marginal hazard ratios with minimal bias. Of these two approaches, IPTW using the propensity score resulted in estimates with lower mean squared error when estimating the effect of treatment in the treated. Stratification on the propensity score and covariate adjustment using the propensity score result in biased estimation of both marginal and conditional hazard ratios. Applied researchers are encouraged to use propensity score matching and IPTW using the propensity score when estimating the relative effect of treatment on time-to-event outcomes.     Raman spectroscopy is a powerful non-destructive technique for qualitatively and quantitatively characterizing materials. However, noise often obscures interesting Raman peaks due to the inherently weak Raman signal, especially in biological samples. In this study, we develop a method based on spectral reconstruction to recover Raman spectra with low signal-to-noise ratio (SNR). The synthesis of narrow-band measurements from low-SNR Raman spectra eliminates the effect of noise by integrating the Raman signal along the wavenumber dimension, which is followed by spectral reconstruction based on Wiener estimation to recover the Raman spectrum with high spectral resolution. Non-negative principal components based filters are used in the synthesis to ensure that most variance contained in the original Raman measurements are retained. A total of 25 agar phantoms and 20 bacteria samples were measured and data were used to validate our method. Four commonly used de-noising methods in Raman spectroscopy, i.e. Savitzky-Golay (SG) algorithm, finite impulse response (FIR) filtration, wavelet transform and factor analysis, were also evaluated on the same set of data in addition to the proposed method for comparison. The proposed method showed the superior accuracy in the recovery of Raman spectra from measurements with extremely low SNR, compared with the four commonly used de-noising methods.

    Determining the foliar N:P ratio provides a tool for understanding nutrient limitation on plant production and consequently for the feeding patterns of herbivores. In order to understand the nutrient limitation at landscape scale, remote sensing techniques offer that opportunity. The objective of this study is to investigate the utility of field spectroscopy and a potential of hyperspectral mapper (HyMap) spectra to estimate foliar N:P ratio. Field spectral measurements were undertaken, and grass samples were collected for foliar N and P extraction. The foliar N:P ratio prediction models were developed using partial least square regression (PLSR) with original spectra and transformed spectra for field and the resampled field spectra to HyMap. Spectral transformations included the continuum removal (CR), water removal (WR), first difference derivative (FD) and log transformation (Log(1/R)). The results showed that CR and WR spectra in combination with PLSR predicted foliar N:P ratio with higher accuracy as compared to FD and R, using field spectra. For HyMap spectral analysis, addition to CR and WR, FD achieved higher estimation accuracy. The performance of FD, CR and WR spectra were attributed to their ability to minimize sensor and water effects on the fresh leaf spectra, respectively. The study demonstrated a potential to predict foliar N:P ratio using field and HyMap simulated spectra and shortwave infrared (SWIR) found to be highly sensitive to foliar N:P ratio. The study recommends the prediction of foliar N:P ratio at landscape level using airborne hyperspectral data and could be used by the resource managers, park managers, farmers and ecologists to understand the feeding patterns, resource selection and distribution of herbivores (i.e. wild and livestock).

    Levels of hardcore drug use have been especially difficult to estimate because of the relative rarity of the behavior, the difficulty of locating hardcore drug users, and the tendency to underreport stigmatized behavior. This chapter presents a new application of ratio estimation, combining sample data from the National Household Survey on Drug Abuse (NHSDA) together with population counts of the number of persons arrested in the past year from the Uniform Crime Report (UCR) and the number of persons in drug treatment programs in the past year from the National Drug and Alcoholism Treatment Unit Survey (NDATUS). The population counts serve as a benchmark accounting for undercoverage and underreporting of hard drug users.

    Non-cancer risk assessment traditionally assumes a threshold of effect, below which there is a negligible risk of an adverse effect. The Agency for Toxic Substances and Disease Registry derives health-based guidance values known as Minimal Risk Levels (MRLs) as estimates of the toxicity threshold for non-carcinogens. Although the definition of an MRL, as well as EPA reference dose values (RfD and RfC), is a level that corresponds to "negligible risk," they represent daily exposure doses or concentrations, not risks. We present a new approach to calculate the risk at exposure to specific doses for chemical mixtures, the assumption in this approach is to assign de minimis risk at the MRL. The assigned risk enables the estimation of parameters in an exponential model, providing a complete dose-response curve for each compound from the chosen point of departure to zero. We estimated parameters for 27 chemicals. The value of k, which determines the shape of the dose-response curve, was moderately insensitive to the choice of the risk at the MRL. The approach presented here allows for the calculation of a risk from a single substance or the combined risk from multiple chemical exposures in a community. The methodology is applicable from point of departure data derived from quantal data, such as data from benchmark dose analyses or from data that can be transformed into probabilities, such as lowest-observed-adverse-effect level. The individual risks are used to calculate risk ratios that can facilitate comparison and cost-benefit analyses of environmental contamination control strategies.

  7. On the Use of Radar Depolarization Ratios for Estimating Shapes of Ice Hydrometeors in Winter Clouds.

    An approach is suggested to relate measurements of radar depolarization ratios and aspect ratios of predominant hydrometeors in nonprecipitating and weakly precipitating layers of winter clouds. The trends of elevation angle dependencies of depolarization ratios are first used to distinguish between columnar-type and plate-type particles. For the established particle type, values of depolarization ratios observed at certain elevation angles, for which the influence of particle orientation is minimal, are then used to estimate aspect ratios when information on particle effective bulk density is assumed or inferred from other measurements. The use of different polarizations, including circular, slant-45° linear, and two elliptical polarizations, is discussed. These two elliptical polarizations are quasi-circular and quasi-linear slant-45° linear, and both are currently achievable with the National Oceanic and Atmospheric Administration Environmental Technology Laboratory's Ka-band radar. In comparison with the true circular and slant-45° linear polarizations, the discussed elliptical polarizations provide a stronger signal in the `weak' radar receiver channel; however, it is at the expense of diminished dynamic range of depolarization ratio variations. For depolarization measurements at the radar elevation angles that do not show much sensitivity to particle orientations, the available quasi-circular polarization provides a better depolarization contrast between nonspherical and spherical particles than does the available quasi-linear slant-45°polarization. The use of the proposed approach is illustrated with the experimental data collected during a recent field experiment. It is shown that it allows successful differentiation among pristine planar crystals, rimed planar crystals, long columns, blocky columns, and graupel.     Abstract-<span class="hlt">Ratios</span> determined from counting a subset of atoms in a sample are positively biased relative to the true <span class="hlt">ratio</span> in the sample (Ogliore et al. 2011). The relative magnitude of the bias is approximately equal to the inverse of the counts in the denominator of the <span class="hlt">ratio</span>. SIMS studies of short-lived radionuclides are particularly subject to the problem of <span class="hlt">ratio</span> bias because the abundance of the daughter element is low, resulting in low count rates. In this paper, we discuss how <span class="hlt">ratio</span> bias propagates through mass-fractionation corrections into an isochron diagram, thereby affecting the inferred initial <span class="hlt">ratio</span> of short-lived radionuclides. The slope of the biased isochron can be either too high or too low, depending on how it is calculated. We then reanalyze a variety of previously published data sets and discuss the extent to which they were affected by <span class="hlt">ratio</span> bias. New, more accurate, results are presented for each study. In some cases, such as for 53Mn-53Cr in pallasite olivines and 60Fe-60Ni in chondrite sulfides, the apparent excesses of radiogenic isotopes originally reported disappear completely. Many of the reported initial 60Fe/56Fe <span class="hlt">ratios</span> for chondrules from ordinary chondrites are no longer resolved from zero, though not all of them. Data for 10Be-10B in CAIs were only slightly affected by bias because of how they were reduced. Most of the data sets were recalculated using the <span class="hlt">ratio</span> of the total counts, which increases the number of counts in the denominator isotope and reduces the bias. However, if the sum of counts is too low, the <span class="hlt">ratio</span> may still be biased and a less-biased <span class="hlt">estimator</span>, such as Beale's <span class="hlt">estimator</span>, must be used. <span class="hlt">Ratio</span> bias must be considered in designing the measurement protocol and reducing the data. A ray-tracing code has been developed to simulate backscattered and depolarized lidar signals from cirrus clouds made of hexagonal-<span class="hlt">based</span> crystals with various compositions and optical depth, taking into account multiple scattering. This code was used first to study the sensitivity of the linear depolarization rate to cloud optical and microphysical properties, then to classify particle shapes in cirrus clouds <span class="hlt">based</span> on depolarization <span class="hlt">ratio</span> measurements. As an example this technique has been applied to lidar measurements from 15 mid-latitude cirrus cloud cases taken in Palaiseau, France. Thus, an experimenter collects measurements consisting of the independent variable (the gas/liquid volume <span class="hlt">ratio</span>) and dependent variable (the GC(-1) peak area). A review of the literature found that the common design is a simple uniform spacing of liquid volumes. Recently, we developed an MR <span class="hlt">ratio</span> imaging technique that approximately yields lipid/{lipid + water}. In this work, we develop a statistical model for the <span class="hlt">ratio</span> distribution that explicitly includes a partial volume (PV) fraction of fat and a mixture of a Rician and multiple Gaussians. Monte Carlo hypothesis testing showed that our model was valid over a wide range of coefficient of variation of the denominator distribution (c.v.: 0-0:20) and correlation coefficient among the numerator and denominator (ρ 0-0.95), which cover the typical values that we found in MRI data sets (c.v.: 0:027-0:063, ρ: 0:50-0:75). Then a maximum a posteriori (MAP) <span class="hlt">estimate</span> for the fat percentage per voxel is proposed. Using a digital phantom with many PV voxels, we found that <span class="hlt">ratio</span> values were not linearly related to PV fat content and that our method accurately described the histogram. In addition, the new method <span class="hlt">estimated</span> the ground truth within +1.6% vs. +43% for an approach using an uncorrected <span class="hlt">ratio</span> image, when we simply threshold the <span class="hlt">ratio</span> image. On the six genetically obese rat data sets, the MAP <span class="hlt">estimate</span> gave total fat volumes of 279 +/- 45mL, values 21% smaller than those from the uncorrected <span class="hlt">ratio</span> images, principally due to the non-linear PV effect.
